IA2AB-ELISA is an accurate enzymatic assay to measure Tyrosine phosphatase (IA2-Ab) Islet Antigen autoantibodies in human serum.
Autoantibodies against IA2-Ab are important pre-clinical markers of type I diabetes mellitus (insulin-dependent diabetes mellitus - IDDM)
- ACCURATE AND RELIABLE ENZYMATIC ASSAY
IA2-Ab molecules are coated onto the ELISA plate wells. IA-2-biotin complexes constitute the liquid phase. IA2-Ab bind to the IA2-Ab coated onto the well, which will also interact with IA-2-biotin forming a bridge. Addition of streptavidin peroxidase (SA-POD) detects the bound IA-2-biotin. The unbound SA-POD in excess is washed off before the addition of the peroxidase substrate, TMB.
Assay procedure: 16-20h at 2-8°C + 1h at 2-8°C + 20 min at RT / shaking + 20 min at RT in the dark.
Working range: 7.5 - 4000 U/mL (NIBSC 97/550)
Sample: serum - 50 µL
Analytical sensitivity: 1.25 U/mL
